51 | Pioneering Skin Longevity Science: A Synergistic Solution to Skin Glycation

Skin glycation, a major driver of aging, occurs when sugar molecules bind to skin proteins such as collagen and elastin, forming Advanced Glycation End-products (AGEs). These AGEs stiffen the skin, reduce elasticity, promote wrinkles, and dull skin tone, representing a critical barrier to skin longevity.
We propose a synergistic anti-glycation solution that integrates inhibition of protein cross-linking, clearance of accumulated AGEs, and modulation of RAGE signaling to prevent downstream cellular damage. This multi-target approach moves beyond conventional single-pathway strategies, offering a comprehensive defense against glycative stress.
A core innovation of our strategy is the incorporation of microRNA (miR-145-5p) identified through transcriptomic screening as a key genetic regulator linking glycation to skin aging. Combined with proteomic profiling, this allowed the identification of functional proteins involved in wrinkle formation (MMP1, MMP3, COL1A1, ELN), inflammation (CXCL8, S100A2), antioxidant defense (SOD2, SIRT3, GPX4), pigmentation (TYRP1, PMEL), and skin barrier integrity (TJP1, KRT16), enabling a precise, multi-level intervention.
To address previously irreversible glycative damage, we developed a novel synergetic combination of soluble RAGE (sRAGE) and autophagy induction, which captures AGEs and blocks downstream damaging signals, effectively restoring protein function.
Clinical evaluation of 190 subjects aged 20–70 over 12 weeks, using a placebo-controlled design, confirmed the solution’s efficacy. Assessment of multiple skin parameters—including ITA° (skin tone), L values (brightness), b values (yellowness), pigmentation, UV chromophores, and AGEs—revealed improvements across six dimensions: anti-glycation, reduced yellowing, brightening, wrinkle prevention, firming, and enhanced elasticity.
In conclusion, this anti-glycation solution is robustly supported by molecular studies, cellular models, and human clinical trials, proving its efficacy in restoring skin youthfulness. It not only provides a comprehensive solution for skin glycation but also contributes a solid scientific foundation for advancing the field of skin longevity science.
